Project Title Antibacterial and Antioxidant properties of Quercetin and its derivatives
Synopsis Dietary flavonoids exhibit exceptional health effects to the human body. Quercetin, a major flavonoid found in fruits and vegetables, has been widely researched. Numerous studies conducted assessed its biological activities, including antioxidant, antibacterial and anti-inflammatory activities. Research was carried out to investigate the effect of methylation and acetylation on the antibacterial and antioxidant activities of the resultant derivatives in order to improve the antioxidant and antibacterial activity of quercetin. In this paper, a convenient method is described, to prepare the derivatives using dimethyl carbonate (DMC) as the reagent for the methylation process, as well as acetic anhydride for the acetylation process. In order to catalyse the reaction, potassium carbonate was used for the methylation process while pyridine was used for the acetylation. The resultant flavonoid derivatives were extracted with high purity using column chromatography. Antioxidant tests were carried out with the DPPH Assay, while antibacterial tests against gram positive B. cereus and gram negative E. coli were conducted using well-diffusion. The tests were carried out for commercial quercetin, methylated quercetin as well as acetylated quercetin. The results of the quercetin derivatives for the assays were compared to the commercial quercetin. In general, methylated quercetin exhibited a decrease in antibacterial and antioxidant properties while acetylated quercetin exhibited a decrease in antioxidant properties while exhibiting an increase in antibacterial properties against B. cereus and a decrease in antibacterial properties against E. coli.
